Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.

Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
=== Professional breastfeeding support === Lactation consultants are trained to assist mothers in preventing and solving breastfeeding difficulties such as sore nipples and low milk supply. They commonly work in hospitals, physician or midwife practices, public health programs, and private practice. Lactation consultants earn their credential, International Board Certified Lactation Consultant (IBCLC), through the International Board of Lactation Consultant Examiners. Breastfeeding support from a lactation consultant is associated with higher rates of any breastfeeding at 6 months but not at 1 month or 3 months post pregnancy based on a meta-analysis of studies conducted in the US and Canada. Peer support for breastfeeding is associated with higher rates of any breastfeeding at 1 month and 3 to 6 months and of exclusive breastfeeding at 1 month, but it is unrelated to breastfeeding outcomes past 6 months post-pregnancy.
Shunting inhibition has no direct effect on the membrane potential of the cell; however, it reduces the effect of any coincident synaptic input by reducing the electrical resistance of the cell's membrane. Shunting inhibition can "override" the excitatory effect of depolarising GABA, resulting in overall inhibition even if the membrane potential becomes less negative. It was thought that a developmental switch in the molecular machinery controlling the concentration of chloride inside the cell changes the functional role of GABA between neonatal and adult stages. As the brain develops into adulthood, GABA's role changes from excitatory to inhibitory.
=== Carcinogens === Soy sauce may contain ethyl carbamate, a Group 2A carcinogen. In 2001, the United Kingdom Food Standards Agency found in testing various soy sauces manufactured in mainland China, Taiwan, Hong Kong, and Thailand (made from hydrolyzed soy protein, rather than being naturally fermented) that 22% of tested samples contained a chemical carcinogen named 3-MCPD (3-monochloropropane-1,2-diol) at levels considerably higher than those deemed safe by the EU. About two-thirds of these samples also contained a second carcinogenic chemical named 1,3-DCP (1,3-dichloropropane-2-ol) which experts advise should not be present at any levels in food. Both chemicals have the potential to cause cancer, and the Agency recommended that the affected products be withdrawn from shelves and avoided. The same carcinogens were found in soy sauces manufactured in Vietnam, causing a food scare in 2007. In Canada, the Canadian Cancer Society writes,
The extent to which the findings of a clinical trial can be reliably extrapolated from the subjects who participated in the trial to a broader patient population and a broader range of clinical settings. (ICH E9) Global assessment variable
===== PI 88788 ===== The vast majority of cultivars in the US have soybean cyst nematode resistance (SCN resistance), but rely on only one breeding line (PI 88788) as their sole source of resistance. (The resistance genes provided by PI 88788, Peking, and PI 90763 were characterized in 1997.) As a result, for example, in 2012 only 18 cultivars out of 807 recommended by the Iowa State University Extension had any ancestry outside of PI 88788. By 2020 the situation was still about the same: Of 849 there were 810 with some ancestry from PI 88788, 35 from Peking, and only 2 from PI 89772. (On the question of exclusively PI 88788 ancestry, that number was not available for 2020.) That was speculated to be in 2012—and was clearly by 2020—producing SCN populations that are virulent on PI 88788.

=== Other applications === In soil science, cation-exchange capacity is the ion-exchange capacity of soil for positively charged ions. Soils can be considered as natural weak cation exchangers. In pollution remediation and geotechnical engineering, ion-exchange capacity determines the swelling capacity of swelling or expansive clay such as montmorillonite, which can be used to "capture" pollutants and charged ions. In planar waveguide manufacturing, ion exchange is used to create the guiding layer of higher index of refraction. Dealkalization, removal of alkali ions from a glass surface. Chemically strengthened glass, produced by exchanging K+ for Na+ in soda glass surfaces using KNO3 melts.

It is an epiphytic and occasionally a lithophytic plant, forming spectacular root bundles. Its cylindric pseudobulbs can grow to a length of 2.5 m. It can grow to gigantic clusters weighing from several hundred kilograms to more than one tonne. One collected in 1893 by Frederick K. Sander & Co. near Penang Island in Malaysia weighed 1 t (0.98 long tons; 1.1 short tons). Half was sent to the Columbian Exhibition in Chicago and the other half to the Singapore Botanic Garden. By 1902, the Singapore specimen had grown to be 14.4 m (47 ft) girth by 3 m (9.8 ft) high, and bore simultaneously 2,090 12.5 cm (4.9 in) wide gold and mahogany-coloured flowers plus 1110 unopened buds. Much more recently, in A.D. 2000, biologists Tim Laman and Phil Atkinson found one in Borneo 7.7 m (25 ft) wide and bearing between 2500 and 5000 flowers. The plant completely encircled the host tree 46 m (151 ft) above the ground. The oldest known individual orchid plant is also a G. speciosum. Planted in the Singapore Botanic Garden in 1861 by Garden Director Lawrence Niven and his staff, it was 154 years old in 2015. The plant is also 5 m (16 ft) in width. Each raceme can grow to a height of 3 m, bearing up to 80 flowers, each 10 cm wide. The flowers are yellow with maroon or dark red spots. These flowers are unusual, since the lowest flowers have no lip and these flowers function as osmophores for the entire inflorescence and continue to emit chemical scent to attract pollinators as flowers open in succession. It blooms only once every two to four years.
Octopuses are highly intelligent. Maze and problem-solving experiments have shown evidence of a memory system that can store both short- and long-term memory. In laboratory experiments, octopuses can readily be trained to distinguish between different shapes and patterns. They have been reported to practise observational learning, although the validity of these findings is contested. The veined octopus collects discarded coconut shells, then uses them to build a shelter, an example of tool use. Octopuses have also been observed in what has been described as play: including moving around a bottle by jetting water at it. Octopuses often break out of aquariums and sometimes into others in search of food. Interpretation of evidence has been used to suggest that octopuses have sentience and can feel pain.
== Post-parliamentary career (2024–present) == In June 2025, Galloway renounced his opposition to Scottish independence, saying that he would now be in favour of a second referendum. Writing on X, he said: “We support the right of the Scots to self-determination. Eleven years after they last did so we believe the time for another referendum is close. Given the collapsing authority of the British state, the pitiful prime minister Starmer and the moral decline of British society, the result cannot easily be predicted. Britain has become a cesspit." This followed a comment Galloway had made on X in September 2024, in which he said: "You COULD persuade me to Scottish Independence, you could never persuade me to the SNP." In August 2025, Galloway announced that he would be contesting the 2026 Scottish Parliament election as the Workers Party candidate for Glasgow Southside. Additionally, he said that he would be second on the party list for the Glasgow region. In a press conference shortly after his election as MP for Rochdale in February 2024, Galloway had said that he was "no longer involved in Scottish politics", though did mention his support of the SNP's stance on Gaza, adding that they had been “outstanding on the Gaza question, at least by comparison with the two big parties of the state”. In light of this, he revealed that friend and lead candidate for Glasgow, Yvonne Ridley, had persuaded him to stand.

Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
Peptides can bind to glass and plastic, so a fraction of the material leaves the solution. The effect is proportionally larger in dilute samples and can bias quantitative results.
Reverse-phase high-performance liquid chromatography is the most widely reported approach. Purity figures should always be quoted together with the wavelength, gradient, and integration parameters used.
