Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.

Reversed-phase high-performance liquid chromatography is widely used to assess purity and to separate the parent peptide from related substances. Mass spectrometry confirms identity and can resolve modifications that differ by a few daltons. Size-exclusion chromatography detects dimers and higher aggregates, which are relevant to both stability and immunogenicity questions. Peptide mapping with enzymatic digestion locates specific modifications along the sequence. Circular dichroism provides a secondary-structure profile, although it gives limited information about local conformational changes.
Quality control for peptide material focuses on identity, purity, content and the profile of impurities. Common degradants include deamidated and oxidised forms, plus aggregates formed during storage or handling. Forced degradation studies under heat, light, acid and peroxide help define which conditions accelerate change and which analytical methods detect it. Limits for individual impurities are set by pharmacopoeial monographs or manufacturer specifications. How much a given impurity affects biological activity is often uncertain, and conclusions may depend on the assay used.
Solid peptide material is generally kept at reduced temperature to limit degradation. Short-term storage at 2 to 8 degrees Celsius is common, while longer archival storage at minus 20 degrees Celsius or below is typical for lyophilised powder. Vials should remain sealed and protected from light, because ultraviolet exposure can oxidise susceptible residues. Repeated freeze-thaw cycles are avoided, as they promote aggregation and loss of soluble material. Solutions are less stable than solids and are usually prepared close to the time of use.
The risk of breast cancer in women with Turner syndrome (45,XO karyotype) also appears to be significantly decreased, though this could be related to ovarian failure and hypogonadism rather than to genetics. Prostate cancer is extremely rare in gonadectomized transgender women who have been treated with estrogens for a prolonged period of time. Whereas as many as 70% of men show prostate cancer by their 80s, only a handful of cases of prostate cancer in transgender women have been reported in the literature. As such, and in accordance with the fact that androgens are responsible for the development of prostate cancer, HRT appears to be highly protective against prostate cancer in transgender women. The risks of certain types of benign brain tumors including meningioma and prolactinoma are increased with hormone therapy in transgender women. These risks have mostly been associated with the use of cyproterone acetate. Estrogens and progestogens can cause prolactinomas, which are benign, prolactin-secreting tumors of the pituitary gland. Milk discharge from the nipples can be a sign of elevated prolactin levels. If a prolactinoma becomes large enough, it can cause visual changes (especially decreased peripheral vision), headaches, depression or other mood changes, dizziness, nausea, vomiting, and symptoms of pituitary failure, like hypothyroidism.
Dasch (1969) showed that surface sediments of Atlantic displayed 87Sr/86Sr ratios that could be regarded as bulk averages of the 87Sr/86Sr ratios of geological terrains from adjacent landmasses. A good example of a fluvial-marine system to which Sr isotope provenance studies have been successfully employed is the River Nile-Mediterranean system. Due to the differing ages of the rocks that constitute the majority of the Blue and White Nile, catchment areas of the changing provenance of sediment reaching the River Nile Delta and East Mediterranean Sea can be discerned through strontium isotopic studies. Such changes are climatically controlled in the Late Quaternary. More recently, 87Sr/86Sr ratios have also been used to determine the source of ancient archaeological materials such as timbers and corn in Chaco Canyon, New Mexico. 87Sr/86Sr ratios in teeth may also be used to track animal migrations. Strontium aluminate is frequently used in glow in the dark toys, as it is chemically and biologically inert.
== Terminology == In the United Kingdom, the name or names of the fruit followed by juice can only legally be used to describe a product that is 100% fruit juice, as required by the Fruit Juices and Fruit Nectars (England) Regulations and the Fruit Juices and Fruit Nectars (Scotland) Regulations 2003. However, a juice made by reconstituting concentrate can be called juice. A product described as fruit "nectar" must contain at least 25% to 50% juice, depending on the fruit. A juice or nectar including concentrate must state that it does. The term "juice drink" is not defined in the Regulations and can be used to describe any drink that includes juice, whatever the amount. Comparable rules apply in all EU member states in their respective languages. In the US, fruit juice can only legally be used to describe a product that is 100% fruit juice. A blend of fruit juice(s) with other ingredients, such as high-fructose corn syrup, is called a juice cocktail or juice drink. According to the Food and Drug Administration (FDA), the term "nectar" is generally accepted in the US and in international trade for a diluted juice to denote a beverage that contains fruit juice or puree, water, and artificial sweeteners. "No added sugar" is commonly printed on the labels of juice containers, but the products may contain large amounts of naturally occurring sugars; however, sugar content is listed with other carbohydrates on labels in many countries.
Directed by Jessie Nelson with musical direction by Lance Horne, the musical was workshopped at the Royal National Theatre and commissioned by the National Theatre Connections. He wrote the music and lyrics to the 2013 musical adaptation of American Psycho, which opened at the Almeida Theatre in London, and was later staged on Broadway in 2016. In 2013, Sheik wrote the music for the musical adaptation of the novel Because of Winn-Dixie, which premiered at the Arkansas Repertory Theatre. Dixie was a collaboration with then-director John Tartaglia and Nell Benjamin, who wrote the book and lyrics. The musical ran at the Goodspeed Opera House in East Haddam, Connecticut from July 2019 to September 2019, directed by John Rando. In 2015, Sheik wrote the musical thriller Noir with Kyle Jarrow. It premiered as part of New York Stage and Film's season in July to August 2015 at the Powerhouse Theater at Vassar College. Inspired by live radio plays and classic film noir, the musical was directed by Rachel Chavkin. In 2016, he prepared the music for the Shakespeare Theatre Company (Washington, D.C.) production of The Taming of the Shrew, described in The Washington Post as "an assortment of preexisting songs by singer-songwriter Duncan Sheik".
==== Sen-chan ==== Senichi Enari (江成 仙一, Enari Sen'ichi), also known as "Sen-chan" (センちゃん), is the eccentric, yet calmest and smartest member, of the team, due in part to his ability to think things through and being able to do his best thinking while in a handstand, which he considers his "thinking pose". Sen-chan serves as Deka Green (デカグリーン, Deka Gurīn). The oldest of seven siblings from a poor family, he displays nycto- and claustrophobia after accidentally falling into a well when he was a child. He was subsequently rescued by a policeman, which inspired him to take up police work. Despite his usually calm demeanor, he is said to be scary when angered. He also displays a crush on his teammate Umeko, but avoids showing it publicly. As of the crossover film Mahō Sentai Magiranger vs. Dekaranger, they have moved in together. As Deka Green, Sen-chan wields the D-Knuckle and D-Rod, which can combine to form the D-Blaster (ディーブラスター, Dī Burasutā) rifle. He also shares riding the Machine Bull (マシンブル, Mashin Buru) police car with Umeko. Using the D-Rod, Sen-chan can perform the Green Crash (グリーンクラッシュ, Gurīn Kurasshu) attack. Sen-chan is portrayed by Yousuke Itou (伊藤 陽佑, Itō Yōsuke). As a child, he is portrayed by Kengo Tajima (田島 健吾, Tajima Kengo).

Subsequently, the Australian government banned pharmacy compounding of GLP-1 medicines, requiring all providers to transition their patients to approved commercial products. In November 2024, Australian regulators and medical experts examined whether advertising by Juniper could indirectly promote prescription-only medicines in breach of the TGA rules. It referred to the use of "medical weight-loss programs" without naming specific drugs, in their advertising. Clinicians and eating-disorder advocates called for stricter oversight of direct-to-consumer marketing in the sector. In 2025, Juniper was among providers whose promotional activity was flagged, including the use of Black Friday sales campaigns and social media engagement. Professional commentary raised concerns about the potential misuse of GLP-1 medications among patients with eating-disorder vulnerabilities. The Australian Health Practitioner Regulation Agency (AHPRA) confirmed that it had taken disciplinary action against individual healthcare practitioners for inappropriate prescribing of GLP-1 receptor agonists to patients, including the issuance of cautions and conditions on registration. During the same period, the TGA requested the removal of more than 3,000 online advertisements for weight-loss medicines, issuing infringement notices for unlawful advertising in September 2025, and releasing updated social-media advertising guidance in November 2025.

Other countries that have expressed interest in OECD membership are Argentina, Brazil, Croatia, Malaysia and Peru. In January 2022, the OECD reported that membership talks were underway with Argentina, Brazil, Bulgaria, Croatia, Peru and Romania. In March 2022, the OECD suspended the participation of Russia and Belarus due to the ongoing Russian invasion of Ukraine.
== Pathogenesis == In susceptible persons, beryllium exposure can lead to a cell-mediated immune response. The T-cells become sensitized to beryllium. Each subsequent exposure leads to an immune response involving CD4+ helper T-lymphocytes and macrophages accumulating in the lungs. As this response continues macrophages, CD4+ T-lymphocytes and plasma cells aggregate together to form the noncaseating granulomas. When beryllium is phagocytized by macrophages, the beryllium triggers macrophage apoptosis, thereby reducing beryllium clearance from the lungs and eventually resulting in secondary necrosis and lysis. Eventually, the outcome is fibrosis of the lung. Several studies have shown that there is a genetic component to beryllium sensitivity. Specifically, those beryllium-exposed workers with a mutation at the HLA-DPB1 Glu69 position have increased prevalence of beryllium sensitization and chronic beryllium disease. The HLA-DPB1 gene is important for MHC class II molecule function on antigen presenting cells. A study of the immune response to beryllium in individuals who express the HLA-DP2 allele found that CD4 T-cells do not detect the Be2+ cation itself, but instead detect surface changes in the HLA-DP2/peptide complex in which Be2+ is embedded. Those researchers concluded that chronic beryllium disease is a predisposition that lies between "allergic hypersensitivity and autoimmunity." According to the International Agency for Research on Cancer, beryllium and beryllium compounds are Category 1 carcinogens; they are carcinogenic to both animals and humans.

=== Biomarkers === ALP enzymes are found abundantly within the bile canaliculi and bile. If a duct is obstructed, tight junctions permit migration of the ALP enzymes until the polarity is reversed and the enzymes are found on the whole of the cell membrane. Serum ALP levels exceeding 2–3 times the upper baseline value may be due to a variety of liver diseases. However, an elevation that exceeds 10 times the upper baseline limit is strongly indicative of either intrahepatic or extrahepatic cholestasis and requires further investigation. Cholestasis can be differentiated from other liver disorders by measuring the proportion of ALP to serum aminotransferases, where a greater proportion indicates a higher likelihood of cholestasis. Typically, aminotransferase enzymes are localized within hepatocytes and leak across the membrane upon damage. However, measurement of serum aminotransferase levels alone is not a good marker to determine cholestasis. In up to a third of patients, ALP levels may be elevated without the presence of cholestasis. As such, other biomarkers should be measured to corroborate findings. Measurement of 5' nucleosidase levels may be used to identify cholestasis in conjunction with ALP. Levels of ALP may rise within a few hours of cholestasis onset while 5' nucleosidase levels may take a few days. Many labs cannot measure 5' nucleosidase and ALP levels so, GGT may be measured in some cases. Abnormal GGT elevation may be attributable to a variety of factors.
Ca(OH)2 + CO2 → CaCO3 + H2O This reaction is slow, because the partial pressure of carbon dioxide in the air is low (~ 0.4 millibar). The carbonation reaction requires that the dry cement be exposed to air, so the slaked lime is a non-hydraulic cement and cannot be used under water. This process is called the lime cycle.
Since the end of the Cold War, coups have become rarer, and more likely to be followed by democratization. Coups still often simply replace one autocracy with another one (with the new autocratic regime usually more repressive, in an attempt to prevent another coup) or have no effect on regime type. As of 2017, there was debate about whether coups in autocracies should now be considered to promote democratization, on average, or if countries' chances of democratization are still unchanged or worsened by coups (since democratization can take place without a coup). One reason for the increase in the chance of democratization is that a higher proportion of coups (half of post-Cold-War coups) now take place in democracies (a higher percentage of countries are also now democracies). Democratic countries often rebound from coups quickly, restoring democracy, but coups in a democracy are a sign of poor political health, and increase the risk of future coups and loss of democracy. The dataset is small, so statistical significance varies depending on the model used, as of 2017; debate will end if data on more coups makes the pattern clear. The post-Cold-War increase in the chances of post-coup democratization may partly be due to the incentives created by international pressure and financing. US law, for instance, automatically cuts off all aid to a country if there is a military coup. According to a 2020 study, "external reactions to coups play important roles in whether coup leaders move toward authoritarianism or democratic governance.

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
Size-exclusion chromatography is the most common technique, separating monomer from dimer and higher-order species by hydrodynamic volume. It is usually paired with ultraviolet detection and, for trace work, with multi-angle light scattering. Results depend on mobile-phase conditions, so method details matter when comparing datasets.
The detailed degradation profile under real-world temperature excursions and repeated handling is not fully described in the open literature. Relative contributions of photo-degradation versus thermal pathways are also incompletely mapped. Whether specific impurity species carry different biological activity is an open question rather than an established finding.
Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
